Receptor tyrosine kinase which binds promiscuously membrane-bound ephrin-A family ligands residing on adjacent cells, leading to contact-dependent bidirectional signaling into neighboring cells. The signaling pathway downstream of the receptor is referred to as forward signaling while the signaling pathway downstream of the ephrin ligand is referred to as reverse signaling. Activated by the ligand ephrin-A1/EFNA1 regulates migration, integrin-mediated adhesion, proliferation and differentiation of cells. Regulates cell adhesion and differentiation through DSG1/desmoglein-1 and inhibition of the ERK1/ERK2 (MAPK3/MAPK1, respectively) signaling pathway. May also participate in UV radiation-induced apoptosis and have a ligand-independent stimulatory effect on chemotactic cell migration. During development, may function in distinctive aspects of pattern formation and subsequently in development of several fetal tissues. Involved for instance in angiogenesis, in early hindbrain development and epithelial proliferation and branching morphogenesis during mammary gland development. Engaged by the ligand ephrin-A5/EFNA5 may regulate lens fiber cells shape and interactions and be important for lens transparency development and maintenance. With ephrin-A2/EFNA2 may play a role in bone remodeling through regulation of osteoclastogenesis and osteoblastogenesis. (Microbial infection) Acts as a receptor for hepatitis C virus (HCV) in hepatocytes and facilitates its cell entry. Mediates HCV entry by promoting the formation of the CD81-CLDN1 receptor complexes that are essential for HCV entry and by enhancing membrane fusion of cells expressing HCV envelope glycoproteins. Acts as a receptor for human cytomegalovirus (HCMV) to mediate viral entry and fusion in glioblastoma cells.

A Phase IB 2 Dose Trial of IRS-1 HSV C134 (IND 17296) Administered Intratumorally in Patients with Recurrent Malignant Glioma

The purpose of this study is to determine how safe and how well-tolerated the experimental study drug, C134 is when administered twice into the brain where the tumor is located. This is a Phase IB 2 dosing study. All the patients who take part in this study will receive the same type of experimental treatment. There is no "placebo" in this study. The patient will receive the dose of C134 administered, which will be added in the tumor infiltrated tissue in the area of the resection cavity. Anywhere from 4-12 patients are expected to take part in the study; the final number will depend on the safety results.

Randomized Phase 2/3 Trial of Vincristine-Irinotecan-Regorafenib in Combination With Vincristine-Doxorubicin-Cyclophosphamide (VDC) and Ifosfamide-Etoposide (IE) in Patients With Newly Diagnosed Metastatic Ewing Sarcoma

This phase II/III trial compares the effect of vincristine, irinotecan, and regorafenib (VIrR) in combination with vincristine, doxorubicin, cyclophosphamide (VDC), ifosfamide and etoposide (IE) to usual treatment with VDC/IE for the treatment of newly diagnosed Ewing sarcoma or other round cell sarcomas that have spread from where they first started (primary site) to other places in the body (metastatic). Vincristine is in a class of medications called vinca alkaloids. It works by stopping tumor cells from growing and dividing and may kill them. Irinotecan is in a class of antineoplastic medications called topoisomerase I inhibitors. It blocks a certain enzyme needed for cell division and deoxyribonucleic acid (DNA) repair and may kill tumor cells. Regorafenib, a type of kinase inhibitor and a type of antiangiogenesis agent, blocks certain proteins, which may help keep tumor cells from growing. It may also prevent the growth of new blood vessels that tumors need to grow. Doxorubicin is in a class of medications called anthracyclines. Doxorubicin damages the cell's DNA and may kill tumor cells. It also blocks a certain enzyme needed for cell division and DNA repair. Cyclophosphamide is in a class of medications called alkylating agents. It works by damaging the cell's DNA and may kill tumor cells. It may also lower the body's immune response. Ifosfamide, a type of alkylating agent and a type of antimetabolite, attaches to DNA in cells and may kill tumor cells. Etoposide is in a class of medications known as podophyllotoxin derivatives. It blocks a certain enzyme needed for cell division and DNA repair and may kill tumor cells. Giving VIrR/VDC/IE may be more effective than usual treatment with VDC/IE in treating patients with newly diagnosed metastatic Ewing sarcoma or other round cell sarcomas.

Mannan is a context-dependent shield that modifies virulence in Nakaseomyces glabratus

Fungal-host interaction outcomes are influenced by how the host recognizes fungal cell wall components. Mannan is a major cell wall carbohydrate and can be a glycoshield that blocks the inner cell wall β-1,3-glucan from activating pro-inflammatory immune responses. Disturbing this glycoshield in Candida albicans results in enhanced antifungal host responses and reduced fungal virulence. However, deletions affecting mannan synthesis can lead to systemic hypervirulence for Nakaseomyces glabratus (formerly Candida glabrata) suggesting that proper mannan architecture dampens virulence for this organism. N. glabratus is the second leading cause of invasive and superficial candidiasis, but little is known about how the cell wall affects N. glabratus pathogenesis. In order to better understand the importance of these species-specific cell wall adaptations in infection, we set out to investigate how the mannan polymerase II complex gene, MNN10, contributes to N. glabratus cell wall architecture, immune recognition, and virulence in reference strains BG2 and CBS138. mnn10Δ cells had thinner inner and outer cell wall layers and elevated mannan, chitin, and β-1,3-glucan exposure compared to wild-type cells. Consistent with these observations, mnn10Δ cells activated the β-1,3-glucan receptor in oral epithelial cells (OECs), EphA2, and caused less OEC damage than wild-type. mnn10Δ replication was also restricted in macrophages compared to wild-type controls. Yet, during systemic infection in Galleria mellonella larvae, mnn10Δ cells induced rapid larval melanization and BG2 mnn10Δ cells killed larvae significantly faster than wild-type. Thus, our data suggest that mannan plays context-dependent roles in N. glabratus pathogenesis, acting as a glycoshield in superficial disease models and modulating virulence during systemic infection.

RSK4 promotes the metastasis of clear cell renal cell carcinoma by activating RUNX1-mediated angiogenesis

Ribosomal S6 protein kinase 4 (RSK4), a member of the serine‒threonine kinase family, plays a vital role in the Ras‒MAPK pathway. This kinase is responsible for managing several cellular activities, including cell growth, proliferation, survival, and mobility. In this study, we observed higher RSK4 protein expression in clear cell renal cell carcinoma (ccRCC) than in normal kidney tissue, and the overexpression of RSK4 might predict poor outcomes for ccRCC patients. Notably, renal cell carcinoma (RCC) is rich in blood vessels; therefore, this study aimed to explore the biological function of RSK4 in ccRCC progression and its specific regulatory mechanism. We analyzed changes in the expression of target genes through transcriptomic and proteomic assessments. We also conducted tube formation assays and VEGF ELISAs to understand the role of RSK4 in angiogenesis. Additionally, we evaluated the regulatory effect of RUNX1 on EPHA2 transcription using a luciferase reporter gene assay and observed that the effect of RUNX1 on activating EPHA2 transcription was negated after the binding site was mutated. Our findings suggested that RSK4 enhanced tube formation by stimulating VEGF secretion. Concurrently, in vivo experiments confirmed that RSK4 expedited RCC metastasis and angiogenesis. This evidence indicates that RSK4 may serve as a new prognostic marker and play a vital role in RCC metastasis.

EphA2 and phosphoantigen-mediated selective killing of medulloblastoma by γδT cells preserves neuronal and stem cell integrity

Medulloblastoma (MB) is a pediatric brain tumor that develops in the cerebellum, representing one of the most common malignant brain cancers in children. Standard treatments include surgery, chemotherapy, and radiation, but despite a 5-y survival rate of approximately 70%, these therapies often lead to significant neurological damage in the developing brain. This underscores the urgent need for less toxic, more effective therapeutic alternatives. Recent advancements in cancer immunotherapy, including immune checkpoint inhibitors and CAR-T cell therapy, have revolutionized cancer treatment. One promising avenue is the use of Gamma Delta (γδ)T cells, a unique T cell population with potential advantages, such as non-alloreactivity, potent tumor cell lysis, and broad antigen recognition. However, their capacity to recognize and target MB cells remains underexplored. To investigate the therapeutic potential of γδT cells against MB, we analyzed the proportion and status of MB-infiltrated γδT cells within patient datasets. We next investigated the expression of γδT cell ligands on MB cells and identified the EphA2 receptor and the phosphoantigen/Butyrophilin complex as key ligands, activating Vγ9 Vδ1 and Vγ9 Vδ2 T cells, respectively, leading to significant MB cell lysis in both monolayer and spheroid models. Importantly, preliminary safety data showed that γδT cells did not target differentiated neurons or neuroepithelial stem cells derived from induced pluripotent stem cells, underscoring the selectivity and safety of this approach. In conclusion, γδT cells trigger an efficient and specific killing of MB and would offer a promising novel therapeutic strategy.

Chen博士。这一消息不禁令人唏嘘，创新药真是十年河东十年河西。 01 十年河东2013 年Daniel Chen与Ira Mellman共同提出了肿瘤免疫循环（Cancer-Immunity Cycle）的概念（图1），这个循环包括七个步骤：1. 癌细胞抗原的释放；2. 癌症抗原的呈递；3. T细胞的启动和激活；4. T细胞向肿瘤的运输；5. T细胞向肿瘤的浸润；6. T细胞对癌细胞的识别；以及7. 癌细胞的杀伤。这7个步骤可分为两个阶段：准备阶段（T细胞应答），主要发生在淋巴结（LNS）；效应阶段（T细胞杀伤），主要发生在肿瘤微环境（TME）。在大多数肿瘤中，癌症免疫循环在这些步骤中的一个或多个被阻断，导致抗癌免疫反应受抑和肿瘤免疫逃逸。图1. Daniel Chen（左）与Ira Mellman（右）十年前的2015年，Keytruda及Opdivo刚刚获批上市，而Tecentriq则刚完成临床3期并取得成功，Tecentriq的临床开发就是由Daniel Chen领导的。肿瘤免疫循环这一概念也逐渐被业内接受，甚至被大多数人奉为圭臬，这也奠定了Daniel Chen与Ira Mellman肿瘤免疫“泰斗”的地位。但随着肿瘤免疫热门靶点，如CD47、Tim3、OX40、4-1BB、CD73、TIGIT，接连在临床上折戟，肿瘤免疫也进入了低谷期。特别是曾被业内寄予厚望的TIGIT单抗临床试验（可以从罗氏临床试验命名SKYSCRAPER看出）的失败直接导致了罗氏制药裁撤了其癌症免疫部门（详见TIGIT的至暗时刻；基因泰克肿瘤免疫之殇），Daniel Chen与Ira Mellman也相继离开了罗氏制药。如今，Daniel Chen成了ADC初创公司的CEO，而Ira Mellman于近日加入了美国Parker Institute for Cancer Immunotherapy (PICI)出任研发总裁，这是一家旨在癌症治疗的科学发现与商业化之间架起桥梁，由世界领先的肿瘤免疫学专家组成的协作联盟的非盈利性质的研究所。如果在十年前，Daniel Chen从罗氏出来创业布局肿瘤免疫，融个几亿美元应该不是难事。而如今Daniel Chen创业选择了ADC赛道从一个侧面暗示：1）肿瘤免疫初创企业已经很难融到钱了；2）Daniel Chen对肿瘤免疫失去了信心；3）ADC仍是一级市场的热点。 02 十年河西2011年及2013年有两款ADC获FDA批准上市，分别是来自Seagen的Adcetris（CD30-MMAE）及Immunogen的Kadcyla（Her2-DM1），掀起了一轮ADC研发热潮（图3）。当时，几乎所有的MNC都与Seagen或Immunogen合作开发ADC药物。图3. FDA批准ADC药物时间线但很快大家发现，以微管抑制剂为Payload的ADC治疗实体瘤的失败率很高，事实上当时推上临床的大部分实体瘤ADC都失败了，比如靶向5T4的ADC（PF-06263507）、靶向EphA2的ADC（MEDI-547）、靶向Mesothelin的ADC（BAY94-9343）等等。这让很多药企很快对ADC丧失了信心，包括罗氏在内的众多MNC纷纷放弃了早期ADC管线。2015年9月，DS-8201进入临床1期试验，那时应该很少有人注意到这个分子，但历史的车轮已经开始悄悄转动。2019年12月，DS-8201在美国获批上市，适应症为Her2阳性乳腺癌。而此后，DS-8201更是一路披荆斩棘，不仅在传统适应症上对Kadcyla形成碾压优势，还开辟了乳腺癌Her2低表达甚至超低表达这些亚型的适应症。而那些具有敏锐嗅觉的中国药企在DS-8201获批上市前就已经开始了Fast follow的脚步并一跃成为ADC行业的领导者，如恒瑞、映恩生物、宜联生物、科伦博泰等等，其ADC管线产品已经成功引起MNC的关注并达成多项合作交易。谁能想到，十年前在ADC领域还默默无闻的第一三共通过消化吸收Seagen的技术结合罗氏的Herceptin及自己在TOPO1i方面的技术积累产生了DS-8201而成为了ADC行业的领导者并成功带领行业实现了跨代升级。再次感叹，创新药真是十年河东十年河西。 03 未来十年未来十年是否会出现一个新的Modality成为市场的新宠，答案是肯定的，但只是也许它还没有浮出水面。近日，阿斯利康/第一三共宣布，DS-8201联合帕妥珠单抗，在HER2阳性乳腺癌一线治疗的三期临床试验中，无进展生存期（PFS）显著优于当前一线标准治疗方案（紫杉烷+曲妥珠单抗+帕妥珠单抗，THP），总生存期（OS）也呈现出积极趋势。DS-8201联合帕妥珠单抗有望成为乳腺癌一线治疗，其市场空间也将进一步放大，甚至可能成为首个百亿美元ADC分子。与此同时，吉利德宣布其靶向Trop-2的ADC药物Trodelvy与默沙东PD-1抑制剂Keytruda联合疗法在 3 期临床试验 ASCENT-04/KEYNOTE-D19 中取得积极的顶线结果。分析显示，该试验达到主要终点，Trodelvy联合Keytruda，与Keytruda联合化疗相比，在临床上显著改善不可手术切除的局部晚期或转移性三阴性乳腺癌（mTNBC）且肿瘤表达 PD-L1（CPS≥10）的患者的无进展生存期（PFS）。未来十年，我们将看到越来越多的ADC与抗体（特别是肿瘤免疫抗体）联合治疗实体瘤进入临床试验并取得成功。我们也将看到更多跨Modality的药物联合治疗多种疾病。共建Biomedical创新生态圈！如何加入BiG会员？

▎药明康德内容团队编辑根据公开资料统计，截至1月31日，2025年1月有十多家专注于创新药研发的中国新锐公司宣布获得新一轮融资。其中至少9家公司融资金额在1亿元及以上，这些获得资本青睐的新锐公司正在开发的产品包括小分子靶向抗肿瘤药物、小型化偶联药物、眼底疾病大分子创新药物、CAR-T细胞治疗药物、分子胶靶向蛋白降解药物等。塔吉瑞生物融资轮次：C轮融资金额：5000万美元 1月22日，塔吉瑞生物宣布完成C轮融资，融资金额5000万美元，本轮融资将用于多个创新药物的国际临床研究的推进。塔吉瑞成立于2014年，专注于新一代小分子靶向抗肿瘤药物研发。根据塔吉瑞官网管线资料，该公司已经搭建了包括6款产品的在研管线，进展较快的产品为第三代ALK抑制剂TGRX-326，拟开发治疗肺癌，目前已经进入中国3期临床研究阶段。2024年9月，先声药业通过一项逾2000万美元首付款的合作获得了TGRX-326在中国大陆的独家商业化权益。塔吉瑞生物其他在研产品还包括Bcr-Abl变构抑制剂TGRX-678，该产品此前已经被中国国家药监局药品审评中心（CDE）纳入突破性治疗品种，拟用于治疗加速期慢性粒细胞白血病（CML-AP）。达歌生物融资轮次：A+轮融资金额：超2000万美元 1月22日，达歌生物（DEGRON THERAPEUTICS）宣布公司于2024年底成功完成了超2000万美元的A+轮融资。本轮融资由浙江省“4+1”生物医药与高端器械产业基金、Takeda Ventures、丰川资本、隆门资本、浩悦中国医疗健康基金以及AIM-HI Accelerator Fund等知名机构投资，现有股东元生创投继续加持。达歌生物专注于通过其专有的基于分子胶的靶向蛋白降解药物发现平台，针对以往无法成药靶点开发新型药物，重点领域包括肿瘤、炎症、神经科学和代谢疾病。根据新闻稿，本轮融资将助力达歌生物在新型分子胶领域的布局，包括推进核心管线HuR分子胶的临床研究。HuR是一种此前被认为无法成药的RNA结合蛋白，在癌症和炎症中发挥关键作用。达歌生物计划于2025年启动临床试验患者招募。星联肽融资轮次：Pre-A+轮融资金额：1亿元 1月16日，星联肽宣布完成1亿元Pre-A+轮融资，本轮融资由复星医药旗下复健资本新药创新基金领投，横琴创投及深圳前海利元共同跟投。星联肽聚焦于小型化偶联药物的研发，包括多肽偶联药物（PDC）和纳米抗体偶联药物（NDC）等。该公司目前已有两款产品处于临床开发阶段，首款产品SC-101（靶向Nectin-4的PDC）分别于2024年初和年中获得中国NMPA和美国FDA的临床默示许可，目前处于1期临床阶段，已初步显示出抗肿瘤效果及良好的安全性。星联肽的第二款产品SC-102（靶向EphA2的PDC）在2024年底进入临床阶段，已完成首例受试者的入组和给药工作。此外，星联肽研发的首个NDC项目有望在今年年中完成PCC提名。阳光安津融资轮次：pre-A轮融资金额：超亿元 1月15日，阳光安津宣布完成超亿元pre-A轮融资。本轮融资由启明创投领投，博行资本和清控银杏跟投。根据阳光安津新闻稿介绍，该公司由雷晓光教授与杨勇教授共同创立，公司以新型镇痛药的研发为目标，专注于Nav1.8和Nav1.7这两个与机体疼痛感受高度相关的钠离子通道。其中，Nav1.8抑制剂已展现了较好的镇痛效果和安全性，该产品预计于2025年启动IND。欧科健生物融资轮次：A轮融资金额：超亿元 1月11日，欧科健生物宣布完成超亿元人民币的A轮融资。此次融资由LongRiver江远投资领投，南山战新投、君实创投、博远资本和清松资本跟投，天使轮股东元生创投持续追投。欧科健成立于2022年8月，专注于眼底常见疾病的大分子创新药物研发，其核心产品OCUL101为一款双靶点三效创新抗体，独特性地针对新生血管性年龄相关性黄斑变性（nAMD）、糖尿病性黄斑水肿（DME）和地图样萎缩（GA）三种适应症，该产品已经相继在美国和中国获批临床。此外，欧科健还针对多种眼底常见疾病进行了完整布局，后续还有多款处于临床前阶段的多抗管线，预计将在2025年底陆续递交中美IND申报。艺妙神州融资轮次：E轮融资金额：数亿元 1月8日，艺妙神州宣布正式完成数亿元人民币的E轮融资。本轮融资由京津冀协同发展产业投资基金领投，太平医疗健康基金、首发展集团、四季青投资、阳光诺和等多家专业机构跟投。根据新闻稿，本轮融资资金将用于支持艺妙神州首款原研CAR-T新药IM19的商业化布局，同时加快其包括血液肿瘤、实体肿瘤以及自身免疫性疾病等重大疾病领域新工艺新产品的研发和临床进程。IM19注射液是一款靶向CD19的自体CAR-T候选产品，其用于治疗复发或难治弥漫大B细胞淋巴瘤（r/r DLBCL）适应症的药品上市许可申请（NDA）已经于2024年11月获得中国NMPA受理。礼邦医药融资轮次：C轮首次融资金额：5.5亿元 1月7日，礼邦医药（Alebund Pharmaceutical）宣布已完成C轮首次5.5亿元人民币融资。本轮投资由某知名产业基金，扬州国金集团、熙诚金睿等共同投资。礼邦医药专注于开发治疗肾脏疾病及相关慢性病创新药物。该公司已经建立了丰富的肾脏病新药产品管线，针对适应症包括慢性肾病（CKD）/透析并发症、IgA肾病、糖尿病肾病、局灶阶段性肾小球硬化、常染色体显性多囊肾病等。根据新闻稿介绍，本轮融资将支持礼邦医药全面推进其肾脏疾病管线的开发和商业化，包括铁基磷结合剂AP301于中国的新药上市申报和全球注册3期临床试验的开展，小分子泛磷转运蛋白抑制剂AP306中美2期临床试验的进一步开展，以及首个商业化产品长效促红细胞生成素在中国的商业化销售网络的扩展。高光制药：融资轮次：Pre-C轮融资金额：1.5亿元 2025年1月3日，高光制药宣布完成1.5亿元人民币的Pre-C轮融资。本轮融资由华金资本、康君资本、英飞尼迪资本共同认购，老股东汉康资本继续加持。高光制药是一家创新药开发公司，专注于研发治疗自身免疫及炎症类疾病、神经退行性疾病的小分子新药。高光制药自主研发的治疗自身免疫疾病和炎症的小分子药物——双靶点TYK2/JAK1抑制剂TLL-018已进入3期注册临床开发阶段，拟开发治疗荨麻疹、银屑病、类风湿关节炎等疾病。此外，高光制药还打造了多个针对神经炎症/中枢神经系统疾病的早期管线，包括具有穿透血脑屏障特性的TYK2/JAK1双抑制剂TLL041，拟开发用于治疗阿尔茨海默病及帕金森病。2023年，Biohaven公司已经与高光制药达成一项约9.7亿美元的合作，获得TLL-041在大中华区域以外的独家开发与许可协议。普祺医药融资轮次：B轮融资金额：超3亿元 2025年1月2日，普祺医药宣布完成了超3亿元B轮系列融资。本轮投资机构包括北京市医药健康产业投资基金、亦庄国投、首发展集团、北科建集团、正奇能科集团等众多机构旗下基金及个人投资者。普祺医药成立于2016年，是一家聚焦免疫/炎症性疾病、以外用药为特色的创新药研发公司，产品管线以皮肤科为主，并涵盖呼吸、眼科等疾病领域。根据新闻稿介绍，本轮融资款项将用于普祺医药现有核心产品普美昔替尼凝胶（PG-011，一款JAK/ROCK双靶点小分子抑制剂）治疗成人及青少年轻中度特应性皮炎的3期临床研究及NDA申报、拓展PG-011在其他适应症的应用及后续其他创新管线的临床推进。此外，该公司拟在北交所进行IPO申报。除了上述公司，本月还有其他多家公司获得新一轮融资，包括：专注于干细胞治疗药物研发的血霁生物获得数千万元新投资；专注于AAV基因疗法研发的金唯科生物完成数千万元阶段性融资；专注于创新型疫苗及新型佐剂研发的简达生物完成近亿元Pre-A轮融资；美杰赛尔获得数千万元的战略投资，推进基因编辑修饰的T细胞、NK细胞以及干细胞等多条产品管线的开发；艾威药业完成A++轮融资，其研发管线涵盖了干眼症、感染性结膜炎、近视眼、青光眼等眼科治疗领域；致力于开发诊疗一体靶向放射性核素疗法的晶核生物完成A+轮近亿元融资；瑞瞳生物完成数千万元天使轮融资，推进眼部长效缓释给药创新产品开发；英百瑞完成近亿元A+轮融资，推进通用型自然杀伤细胞疗法的临床研发；安炎达医药完成数千万元人民币Pre-A轮融资，该公司聚焦于开发针对免疫炎症领域的创新药等等。期待在资本的助力下，更多前沿疗法和技术能够更快惠及患者。参考资料： [1]各公司官网及公开资料本文来自药明康德内容团队，欢迎个人转发至朋友圈，谢绝媒体或机构未经授权以任何形式转载至其他平台。转载授权及其他合作需求，请联系wuxi_media@wuxiapptec.com。免责声明：药明康德内容团队专注介绍全球生物医药健康研究进展。本文仅作信息交流之目的，文中观点不代表药明康德立场，亦不代表药明康德支持或反对文中观点。本文也不是治疗方案推荐。如需获得治疗方案指导，请前往正规医院就诊。
