Catalyzes the first and rate-limiting reaction of the four reactions that constitute the long-chain fatty acids elongation cycle (PubMed:29496980, PubMed:30487246). This endoplasmic reticulum-bound enzymatic process allows the addition of 2 carbons to the chain of long- and very long-chain fatty acids (VLCFAs) per cycle. Condensing enzyme that exhibits activity toward saturated and monounsaturated acyl-CoA substrates, with the highest activity towards C22:0 acyl-CoA. May participate in the production of both saturated and monounsaturated VLCFAs of different chain lengths that are involved in multiple biological processes as precursors of membrane lipids and lipid mediators. Important for saturated C24:0 and monounsaturated C24:1 sphingolipid synthesis (PubMed:20937905). Indirectly inhibits RPE65 via production of VLCFAs.

Relationship between time-dependent epidermal ceramide composition changes and skin barrier function in adult mice

Ceramides, especially acylceramides and protein-bound ceramides, are important for skin barrier formation. However, due to the neonatal lethality of knockout (KO) of the genes involved in the production of these ceramides, the effects of their KO in adult mice have been unclear. To investigate these effects, we created mice with tamoxifen-inducible conditional KO of the fatty acid elongase Elovl1. Following tamoxifen administration, acylceramide levels began to decrease from day 5. On day 10, impaired formation of lipid lamellae and thickening of the epidermis were observed. On day 15, protein-bound ceramide levels were substantially reduced and transepidermal water loss was increased. Changes in quantities of ceramides other than acylceramides and protein-bound ceramides and shortening of their fatty acid moieties were also observed, but time courses differed among ceramide classes. RNA sequencing revealed changes in the expression levels of genes involved in ceramide metabolism and keratinocyte proliferation and differentiation in Elovl1 conditional-KO mice. In summary, this study reveals that acylceramides and protein-bound ceramides are important for maintaining the skin barrier in adults, although they are not essential for survival. We also observed compensatory responses toward reduced skin barrier function, such as changes in gene expression, epidermal morphology, and ceramide composition.

ELF5 gene promotes milk lipid synthesis in goat mammary epithelial cells by transcriptomic analysis

E74-like factor 5 (ELF5) is an Ets transcription factor of epithelial development, while the function of ELF5 gene in goat milk fat synthesis remains to be elucidated. In goat mammary epithelial cells, we performed RNA sequencing and analyzed differentially expressed genes (DEGs) after ELF5 gene overexpression. ELF5 gene significantly up-regulated the synthesis of triglyceride, total cholesterol, free fatty acid, and lipid droplets. We obtained 929 DEGs after ELF5 gene overexpression in GMECs. Among the DEGs, SPP1, ELOVL1, PNPLA2, FOXO1, PTGS2, SEMA6A, ACSL5, and GPNMB genes that are related to lipid metabolism were identified. Enrichment analysis showed MAPK and FoxO signaling pathways were up-regulated by ELF5 gene overexpression in GMECs. These findings offer evidence that ELF5 gene could be a candidate gene for the regulation of milk lipid synthesis in goats, and provide molecular targets for the breeding of goats with high milk fat.

Changes in the Expression of Some Genes With Metabolic, VLDL and Antioxidative Effects After the Addition of Essential Oil Mixture to Drinking Water in the Liver of Domestic Geese (Anser anser Domesticus)

ABSTRACTStudies have shown that essential oils not only increase cell viability but also affect lipid metabolism in mammals. However, the extent to which these effects are realized in goose liver has not yet been fully elucidated. The object of research is to investigate the effects of four essential oil mixtures (juniper oil, mint oil, thyme oil, rosemary oil) on lipid metabolic gene expressions in goose. We measured mRNA levels of metabolic genes (ACSBG2, ELOVL1, ELOVL2, CYP2Cl9, CYP2K1), antioxidative gene (SOD1) and very low‐density lipoprotein triglyceride (VLDL) synthesis genes (APOB, FOXO1, MTTP), in goose (Anser anser) liver. Search groups were formed as C (control; no additives), EK1 (0.4 mL/L essential oil mixture supplemented) and EK2 (0.8 mL/L essential oil mixture supplemented). The relative expression levels of genes in the liver were measured using RT‐qPCR. β‐Actin was used as reference gene control for normalization of qPCR data. As a result, essential oil supplementation downregulated metabolic genes compared to the control group. APOB gene among VLDL genes was significantly downregulated. Antioxidative effect gene was downregulated in parallel with the others. This indicates that essential oil intake with drinking water downregulates the genes involved in lipid metabolism in goose liver. Our data show that essential oils have a significant effect on the regulation of genes and pathways involved in hepatic lipid metabolism.
