Catalyzes the oxidative deamination of primary amines to the corresponding aldehydes with the concomitant production of hydrogen peroxide and ammonia (PubMed:19588076, PubMed:24304424, PubMed:9653080). Has a preference for the primary monoamines methylamine and benzylamine (PubMed:19588076, PubMed:9653080). Could also act on 2-phenylethylamine but much less efficiently (PubMed:19588076). At endothelial cells surface can also function as a cell adhesion protein that participates in lymphocyte extravasation and recirculation by mediating the binding of lymphocytes to peripheral lymph node vascular endothelial cells in an L-selectin-independent fashion (PubMed:9254657, PubMed:9653080). Has no semicarbazide-sensitive amine oxidase (SSAO) activity.

The Oncogenic role of Lysyl Oxidase-Like 1 (LOXL1): Insights into cancer progression and therapeutic potential

Lysyl oxidase-like-1 (LOXL1) is a copper-dependent amine oxidase that maintains the structural integrity of the extracellular matrix (ECM) by catalyzing the cross-linking of collagen and elastin. However, aberrations in LOXL1 expression can contribute to diseases like glaucoma, tissue fibrosis, and cancer. LOXL1 has been found to be overexpressed in various malignancies, playing a pivotal role in tumor growth and metastasis. Although some studies suggest tumor-suppressive attributes of LOXL1, its role in tumorigenesis remains controversial. Research on LOXL1 has been primarily focused on pseudoexfoliation syndrome/glaucoma, with limited reviews on its impact on cancer. This review aims to explore LOXL1 comprehensively, including its structure, biological effects, and regulatory processes. Emphasis is placed on understanding the relationship between LOXL1 and tumorigenesis, specifically how LOXL1 influences tumor microenvironment remodeling, tumorigenesis, and metastasis. The review also discusses potential therapeutic strategies targeting LOXL1 for anti-fibrosis and anti-tumor interventions.

Ectopic Overexpression of Rapeseed BnaNTL1 Transcription Factor Positively Regulates Plant Resistance to Sclerotinia sclerotiorum through Modulating JA Synthesis and ROS Accumulation

Oilseed rape is one of the most important oil crops worldwide. Stem rot disease of rapeseed is caused by Sclerotinia sclerotiorum, posing a threat to oil crop yield. NTL is a small class of membrane-tethered NAC transcription factors, which are stored on the membrane in dormant form, released upon stimuli, and then transported to the nucleus. Here, we identified BnaNTL1 from oilseed rape, and its relocation from the ER to the nucleus is induced by S. sclerotiorum. Plants overexpressing BnaNTL1-ΔTM (a truncated form without the transmembrane domain) are resistant to S. sclerotiorum infection and are accumulated with more JA and ROS. Genes related to the JA pathway and ROS signal were significantly induced by BnaNTL1. Furthermore, the dual-luciferase and EMSA results showed that BnaNTL1-ΔTM directly binds to the promoter regions of AOC3, LOX2, OPCL1, and PDF1.2, and it activates their expression. In summary, we identified that BnaNTL1 positively regulates plant resistance to S. sclerotiorum infection by modulating JA synthesis and ROS production.

Cardiovascular risk prediction in hemodialysis patients using the triglyceride-glucose index: a multicenter prospective cohort study

ABSTRACTBackgroundTriglyceride-glucose (TyG) index has recently been established as an indicator of insulin resistance and has predictive value for cardiovascular (CV) disease. However, the clinical significance of the TyG index in patients undergoing hemodialysis remains unknown.MethodsWe prospectively enrolled 759 patients undergoing maintenance hemodialysis. The participants were divided into tertiles based on their baseline TyG index. Echocardiographic parameters, vascular calcification scores, and several plasma biomarkers were obtained and compared using the TyG index.ResultsThe TyG index was positively correlated with levels of circulating vascular pathologic markers, endostatin (ρ = 0.134, P = .025) and vascular adhesion protein-1 (ρ = 0.130, P = .012), but not with vascular calcification score. The TyG index was not correlated with any echocardiographic parameters. Patients in tertile 3 showed the highest cumulative event rates of CV and cardiac events (P < .001 and P = .001, respectively). In the multivariable Cox regression analysis, patients in the TyG index tertile 3 had a significantly increased risk of CV and cardiac events compared to those in the TyG index tertile 1 [adjusted hazard ratio (HR): 1.89, 95% confidence interval (CI): 1.08–3.30, and adjusted HR: 2.01, 95% CI: 1.05–3.82, respectively]. A 1 standard deviation increase in the TyG index was also associated with significantly higher risks of CV and cardiac events.ConclusionsThe TyG index was associated with vascular pathology markers and an increased risk of adverse CV outcomes in patients undergoing hemodialysis. Our study suggests that the TyG index has the potential to assist clinicians in identifying a high CV risk in hemodialysis patients.

STTT：中山大学徐瑞华/鞠怀强团队发现，ACAT1会召集自然杀伤细胞对抗肠癌！

*仅供医学专业人士阅读参考我们知道，微卫星稳定型结直肠癌（CRC）是典型的“冷”肿瘤。这类肿瘤通常对免疫疗法反应较差，且肿瘤微环境也呈现免疫抑制状态。因此，找到改善CRC肿瘤微环境的方法，是将“冷”肿瘤转化为“热”肿瘤，克服CRC免疫治疗难题的关键。自然杀伤（NK）细胞作为先天免疫系统的关键杀伤细胞，能在不依赖抗原呈递的情况下有效识别并杀伤肿瘤细胞。既往有研究显示，肿瘤中NK细胞浸润与CRC患者预后呈正相关。不过，NK细胞在肿瘤微环境中的功能和数量可能会受到肿瘤代谢（比如营养竞争、代谢酶相关免疫信号通路调控）的影响。但截至目前，肿瘤代谢对NK细胞功能的具体影响还不清楚。近期，中山大学徐瑞华/鞠怀强团队就发表了一项重要研究成果。他们发现，线粒体代谢酶ACAT1可通过核转位的方式促进肿瘤微环境中NK细胞的浸润和激活，从而增强NK细胞的抗肿瘤免疫反应。具体来说，通过对CRC患者的多组学与生存数据进行系统性分析，研究人员先是发现，线粒体代谢酶ACAT1的表达水平与NK细胞浸润程度呈正相关。随后的进一步研究发现，在免疫刺激（如IL-18）作用下，原本定位在线粒体中的ACAT1，会在丝氨酸60（S60）位点发生磷酸化，并转移到细胞核中，发挥乙酰转移酶活性，且能在细胞核中特异性乙酰化NF-κB家族成员p50的赖氨酸146（K146）位点，进而削弱其DNA结合能力和转录抑制功能，激活多个免疫相关因子（如CCL5、CXCL10）的表达，这进一步促进了NK细胞的募集和活化，增强了NK细胞对CRC细胞的杀伤能力，从而抑制癌症进展。此外，研究还发现，营养匮乏的肿瘤微环境因素会抑制ACAT1的核转位过程，而核内ACAT1 S60磷酸化水平下降与NK细胞浸润减少、预后不良密切相关。研究发表在Signal Transduction and Targeted Therapy上[1]。为了探讨在CRC中，肿瘤代谢对NK细胞功能的具体影响。研究人员对CRC患者的多组学数据和生存数据进行了系统分析，并排查了与促进NK细胞浸润的代谢相关蛋白。根据分析结果，排在前3名的分别是谷胱甘肽S-转移酶mu2（GSTM2）和铜胺氧化酶3（AOC3），以及线粒体乙酰辅酶A乙酰转移酶1（ACAT1），其中前两种酶的免疫功能已被广泛报道，因此研究人员主要关注了ACAT1。接下来通过进一步验证，研究人员发现，ACAT1蛋白表达水平与1-2期CRC患者肿瘤微环境中NK细胞浸润程度呈正相关。且ACAT1高表达与也CRC患者的总生存率更高有关。随后在构建的CRC小鼠模型中，研究人员发现，只有在免疫系统正常的小鼠体内，ACAT1才能显著抑制肿瘤生长，提示ACAT1的抗肿瘤作用依赖免疫系统。而进一步的流式细胞术等一系列试验证实，ACAT1能够增加肿瘤组织内的NK细胞数量及其活性（表现为增加了NK细胞介导的细胞毒性基因，如IFNG,RAC1的表达）。鉴于ACAT1不仅可以在线粒体内发挥作用，也可以在细胞核中发挥作用，于是，通过构建带有核内定位信号（NLS）的ACAT1和核外输出信号（NES）的ACAT1，研究人员发现，只有NLS标记的ACAT1能够促进肿瘤组织中NK细胞的数量，同时有效抑制肿瘤生长。进一步，利用质谱分析和免疫沉淀实验，研究人员揭示了核内ACAT1促进NK细胞在肿瘤微环境中浸润的机制。即在免疫刺激（如IL-18）作用下，原本定位在线粒体中的ACAT1，会在S60位点发生磷酸化，并转移到细胞核中，发挥乙酰转移酶活性，且能在细胞核中特异性乙酰化NF-κB家族成员p50的赖氨酸146（K146）位点，进而削弱其DNA结合能力和转录抑制功能，激活多个免疫相关因子（如CCL5、CXCL10），以及其他与NK细胞活化有关的分子（如HLA-C、ICAM3、ULBP1和ULBP2）的表达，这进一步促进了NK细胞的募集和活化，从而增强了NK细胞对CRC细胞的杀伤能力，从而抑制癌症进展。此外，研究还发现，营养匮乏的肿瘤微环境因素会抑制ACAT1的核转位过程，而通过对CRC患者的肿瘤组织进行免疫荧光和免疫组化分析，研究人员发现，核内ACAT1 S60磷酸化水平下降与NK细胞浸润减少、预后不良密切相关。总之，本研究揭示了，ACAT1通过其核内转位和对p50 K146的乙酰化，增强了NK细胞的浸润和激活，从而抑制CRC的生长的机制。而鉴于ACAT1 S60磷酸化水平与NK细胞浸润程度及患者的预后密切相关，提示，未来核内ACAT1 S60磷酸化水平可能成为CRC预后的一个重要生物标志物。同时，这些结果也为开发新的CRC免疫治疗方案提供了理论依据。参考文献：[1]Wei C,Liao K,Chen HJ,et al.Nuclear mitochondrial acetyl-CoA acetyltransferase 1 orchestrates natural killer cell-dependent antitumor immunity in colorectal cancer.Signal Transduct Target Ther.2025;10(1):138.Published 2025 Apr 28.doi:10.1038/s41392-025-02221-y本文作者丨张金旭
