Plays a role as a central modulator of cardiovascular and renal function and nociception. Also plays a role in energy metabolism and storage. Inhibits adrenocortical cell proliferation with minor stimulation on corticosteroid release (By similarity). Acts as a ligand for galanin receptors GALR2 and GALR3 (PubMed:17284679, PubMed:24517231). Intracerebroventricular administration of the peptide induces an increase in arterial blood pressure, a decrease in both heart rate and renal excretion and delayed natriuresis. Intraventricular administration of the peptide induces antinociceptive activity. Also induces contraction of muscarinic-like stomach smooth muscles. Intraperitoneal administration of the peptide induces a reduction in food consumption and body weight. Inhibits long chain fatty acid uptake into adipocytes (By similarity). Intracerebroventricular administration of the peptide induces a decrease in heart rate, but no change in arterial pressure, and an increase in urine flow rate. Intraventricular administration of the peptide induces antinociceptive activity (By similarity).

Spexin expression in the human bile duct and perihilar cholangiocarcinoma

The bile duct transports bile fluid from the liver to the gallbladder and small intestine. It contains bioactive peptides, including galanin (GAL) and its receptors (GAL_1-3-R). Spexin (SPX), a member of the GAL peptide family, activates GAL₂-R and GAL₃-R. Its expression in perihilar bile ducts or in perihilar cholangiocarcinoma (pCCA), the most common biliary cancer, is largely unknown. This study investigated SPX expression in healthy, cholestatic, and malignant bile duct tissues. Immunohistochemistry was used to evaluate SPX in healthy (n = 4), peritumoral (PIT) (n = 23) and pCCA (n = 34) tissues. Score values of SPX expression were calculated and statistically analyzed. In healthy and PIT tissues with or without cholestasis, SPX expression was predominantly observed in cholangiocytes and nerve fibers. In pCCA, tumor cells also expressed SPX. SPX levels were similar across healthy, peritumoral, and cholangiocytes/tumor cells. In a small pCCA patient cohort (n = 19), SPX expression did not correlate with tumor grade or patient survival (p = 0.0838). The substantial expression of SPX in cholangiocytes and nerve fibers in the bile duct indicates that SPX contributes via galaninergic signaling to gall bladder function. The presence of SPX in submucosal nerve fibers suggests a neuromodulatory role, possibly involving bile duct motility. SPX expression did not correlate with survival in pCCA, whereas previous findings on GAL suggest a prognostic value. This highlights the need for joint studies of SPX and GAL in larger cohorts.

High relative humidity mitigates the adverse effects of excessive light by protecting photosynthetic machinery in flowering Chinese cabbage

Increasing canopy relative air humidity (RH) facilitates the resistance of flowering Chinese cabbage plants against excessive light exposure in a plant factory with artificial lighting (PFAL), thereby completely inhibiting leaf burn occurrence. To clarify this high RH-mediated resistance mechanism, we further analyzed the transcriptomes, gas exchange parameters, and chlorophyll fluorescence of flowering Chinese cabbage plants subjected to two levels of canopy RH (70% and 90%). Transcriptomic data revealed a significant enrichment of photosynthesis antenna proteins pathway, which was notably downregulated in both the cotyledons and the first true leaves of the plants grown at 70% RH from the 10th to the 14th day after sowing. However, the downregulation of the photosynthesis-antenna proteins pathway in the first true leaves was notably attenuated by increasing the RH from 70% to 90%, consequently inhibiting the down-regulated expression of all light-harvesting complex I- and II-subunit-encoding mRNAs. The 70% RH-treated seedlings exhibited serious photoinhibition, as indicated by relatively lower non-photochemical quenching (NPQ), maximal quantum yield of photosystem I and II, and adaptation of the photosynthetic apparatus to high irradiance. The 90% RH treatment greatly lessened the reduction of NPQ and relative electron transport rates, which favored dissipating the excess excited energy, protected the photosynthetic apparatus against photodamage, and ultimately completely inhibited leaf burn occurrence. Overall, our results indicate that high canopy RH could improve the resistance of plants to intense light by maintaining efficient photosynthesis, thereby minimizing reliance on supplemental artificial lighting.

Antarctic lichens exhibit diverse photobiont distributions and a complex regulation of non-photochemical quenching

The structure, function, and molecular mechanisms of lichen survival in harsh habitats like Antarctica and Alpine localities, where environmental extremes change frequently, are highly interesting yet largely underexplored. We used high resolution microscopy, Raman spectroscopy, and chlorophyll a fluorescence to investigate the basic structure and function, i.e., intrathalline distribution and allocation of photobionts, as well as the heat dissipation process in three Antarctic lichens: Dermatocarpon polyphyllizum (DP), Umbilicaria antarctica (UA), and Leptogium puberulum (LP). Microscopic images of their transverse slices revealed visual insights into the heterogeneous distribution of photobionts within their structurally distinct thalli. Raman spectra showed shifts in the carotenoid Raman ν₁(CC) band between lichens with algal (DP and UA) and cyanobacterial (LP) photobionts, and interestingly, they revealed biosynthesis of scytonemin, a UV-screening pigment, in cyanolichen LP. We found that increasing actinic irradiance has a nearly equal effect on the shape of chlorophyll fluorescence transients also during dark relaxation in lichens with algal photobionts, but it differed greatly for cyanolichen LP. The dark relaxation kinetics of non-photochemical quenching (NPQ) in experimental lichens differed significantly between lichens with algal photobionts DP and UA; however, this parameter could not be calculated in cyanolichen LP. The components of NPQ revealed that rapidly relaxing energy dependent quenching, Φ_qE, is active and protects the thallus of DP predominantly; however, in UA state transition quenching, Φ_qT, predominates. The diversity in NPQ across the three examined lichens revealed intriguing aspects of heat dissipation in their photobionts as a mechanism for survival under Antarctica conditions.

Nature | 姜道华团队揭示磷酸转运蛋白XPR1转运和调控的分子机制
